As shown in the following figure, D depicts the fluorescent cells which were analyzed. Zebrafish transient microinjections result was in a mosaic expression; therefore, only some cells expressed the GCaMP. A red fluorescent transgene was co-expressed as background control. In B, which is the data from the 3dpf fish, each in a different color. Cells were individually measured and combined as mean +/- SEM. The ration of Green (GCaMP fluorescence) was calculated at 488nm: 561nm (dsRed fluorescence). Based on the fluorescence ratios there were spikes at 3 days post fertilization(dpf) in the fish after the treatment concentration is increased. The result ware consistent with the expectation. After analyzing the data, there seemed to be a clear response at sac 0.5%, 1%, 1.5%, 2% within 2-3min.
